Children's Hospital of Philadelphia researcher leads largest-ever genome-wide study of this pediatric condition
Genetics researchers have identified at least two new gene variants that increase the risk of common childhood obesity.
"This is the largest-ever genome-wide study of common childhood obesity, in contrast to previous studies that have focused on more extreme forms of obesity primarily connected with rare disease syndromes," said lead investigator Struan F.A. Grant, Ph.D., associate director of the Center for Applied Genomics at The Children's Hospital of Philadelphia. "As a consequence, we have definitively identified and characterized a genetic predisposition to common childhood obesity."
The study, by an international collaborative group, the Early Growth Genetics (EGG) Consortium, appeared online today in Nature Genetics.
As one of the major health issues affecting modern societies, obesity has increasingly received public attention, especially given a rising prevalence of the condition among children. Research indicates that obese adolescents tend to have higher risk of mortality as adults. Although environmental factors, such as food choices and sedentary habits, contribute to the increasing rates of obesity in childhood, twin studies and other family-based evidence have suggested a genetic component to the disease as well.
Previous studies have identified gene variants contributing to obesity in adults and in children with extreme obesity, but relatively little is known about genes implicated in regular childhood obesity.
"The Center for Applied Genomics at the Children's Hospital of Philadelphia has recruited and genotyped the world's largest collection of DNA from children with common obesity," said Grant. "However, in order to have sufficient statistical power to detect novel genetic signals, we needed to form a large international consortium to combine results from similar datasets from around the world."
The National Institutes of Health partly funded this research, which analyzed previous studies supported by many other European, Australian and North American organizations.
The current meta-analysis included 14 previous studies encompassing 5,530 cases of childhood obesity and 8,300 control subjects, all of European ancestry. The study team identified two novel loci, one near the OLFM4 gene on chromosome 13, the other within the HOXB5 gene on chromosome 17. They also found a degree of evidence for two other gene variants. None of the genes were previously implicated in obesity. "The known biology of three of the genes," added Grant, "hints at a role of the intestine, although their precise functional role in obesity is currently unknown."
"This work opens up new avenues to explore the genetics of common childhood obesity," said Grant. "Much work remains to be done, but these findings may ultimately be useful in helping to design future preventive interventions and treatments for children, based on their individual genomes."

The co-first author of the paper, Jonathan P. Bradfield, is from The Children's Hospital of Philadelphia. Two senior investigators from Children's Hospital, Hakon Hakonarson, M.D., Ph.D., director of the Hospital's Center for Applied Genomics, and Robert I. Berkowitz, M.D., director of the Weight and Eating Disorders Research Program, were also among the study co-authors.

For a music superstar who has several personas living in her creative genius, it almost makes sense that Nicki Minaj should defy all logic when it comes to musical genre.
When she dropped her album Pink Friday: Roman Reloaded this week, she set herself up for two career tracks — one being Nicki Minaj, the tough Queens rapper who spits lines like snakes spit venom, and the other being Nicki Minaj, the colorful, sassy pop songstress who wants to make you pump your fists. So, does this mean she has reloaded herself as a pop star as well as MC and picked up where "Super Bass" left off?
Being that her whole persona is built on being colorful, loud, feisty, brash and fashionable, she's the perfect combination of Lady Gaga, Rihanna, Britney Spears and, yes, Lil Kim, paving her way for what looks to be a career full of success. The kind of success that can satisfy urban radio, which supported her from her grimy mixtape days, but also satiate the appetites of her fans who loved when she blasted "Super Bass" on the Femme Fatale Tour alongside Spears.
She's a conundrum. She exists in two worlds and seems unapologetic about it. Roman Reloaded toes the line, thanks to hip-hop production creds from Hitboy and Pop and Oak — who created hard-rap tracks — and then she gets schizo with RedOne and Dr. Luke, making really, really big pop records.
The pop songs work, but given that Nicki has spent so much more time as a rapper, she doesn't seem to have found her pop voice just yet. They are, at times, mediocre. Her ability to play different characters on the rap songs is what makes her stand out and makes her so engaging. She's a dude one moment, British another, some other combination of gender and nationality the next. So it feels like finding a pop persona should come a bit easier for her.
While she might be wavering at times sonically — trying to nail down what her pop imprint will sound like — visually, she looks like a pop star. She wears colorful wigs, body-hugging couture and costumes that unless you're a pop star or it's Halloween, you could never get away with it. Between the music and the look, she's already a foot and a half in the pop door.
Reloaded is her first full entry into the world of pop: It's confessional at times ("Marilyn Monroe") and just plain fun others ("Starships"), and somewhere in there lies "Nicki: The Pop Star." She's not fully realized yet, but she's bucking the system. And much like Nicki's persona Roman, she's aiming to break free from her hip-hop boarding school and play in the pop big leagues. Let's just hope she never forgets where she came from.
She's definitely a femme fatale, like her pal Britney, and on her way to pop mega-stardom.

Why 49% of preschoolers don't go outside to play | BabyCenter
Does your preschooler go outside to play at least once a day? Maybe not.
According to a new study published in the Archives of Pediatric & Adolescent Medicine, about half of participating preschoolers didn?t go outside at least once per day with their parents.
The study, conducted by Dr. Pooja Tandon and colleagues, targeted almost 9000 children, a nationally representative sample of preschool-aged kids in the United States. Although 93% of the mothers in this sample said their neighborhood was safe, only 44% of mothers and 24% of fathers reported taking their children outside at least once a day.
As you might expect, kids with working mothers were slightly less likely to get outside. And the children of parents who exercise regularly got out more often.
There were also results that you might not have predicted. Despite those daycare playgrounds, kids in childcare were less likely to get outside each day. So were girls and children representing ethnic minorities. You can hear Dr. Tandon discuss the results of the study here:
As Pooja Tandon notes elswhere, this is all very worrying. ?Research shows that outdoor time is important for children?s physical activity and also beneficial for their motor skills, learning, vitamin D levels, vision, and mental health.?
But as a parent herself, she knows it can be difficult to find the time for supervised, outdoor play. The solution? She makes several suggestions:
? Find out if your child gets outside at school or in daycare, and advocate for the importance of daily outdoor play.
? Re-think your habits and see what family activities can be moved outside. Can you walk instead of drive? Turn indoor meals into outdoor picnics? Arrange play dates at your local park?
? Don?t let ?bad? weather keep you inside. That?s what raingear and winter clothing is for.
? Is it dark by the time you get home from work? Try going on family walk with flashlights.
These suggestions sound good to me. But as an anthropologist whose spent a lot of time thinking about the evolution of childhood, I?m struck by another point:
What really ensures that young children play outside is the presence of other kids.
It used to be a universal of childhood. Children living in hunter-gatherer bands or traditional agricultural villages didn?t live indoors. They spent their days outside, in mixed-aged playgroups, the older children looking after the younger ones.
Adults didn?t supervise or chaperone, though the children weren?t completely on their own, either. Somebody ? a parent or friendly neighbor ? was usually within earshot, and available should an emergency arise. Children played on what anthropologist David Lancy calls the ?mother ground,? a village green or commons.
And please note, in case you missed it:
Parents didn?t play with their children, or arrange play dates, or create toddler-friendly outdoor activities.
Adults were too busy ? and children too independent ? for that.
So I can?t help thinking. Surely it?s the loss of the ?mother ground? ? and the close-knit, family-oriented community that comes with it ? that explains why so many toddlers in Tandon?s study don?t get outside.
American parents ? and their counterparts in other countries ? aren?t lazy. It?s not as if we are shirking duties that our parents and grandparents performed. What?s changed is that parents are now expected to supervise their children whenever they go outside.
So there is a conflict ? a new conflict. Children?s outdoor time versus the parents? need to work indoors. Can we fix that problem? Are there tips for parents to re-create the sort of community where children play with other children ? outside ? on the mother ground? Perhaps so. But it will depend on lot of cooperation between adults. What we really need is a village.
